After The Detector
Following the detector, the flow path often leads
to the waste reservoir for completion of the basic HPLC system. One
nice accessory to place inline along the flow path is a 100 psi Back
Pressure Regulator, our part number P-787.
This is usually sufficient pressure to prevent outgassing within your
flow cell, but not so high as to damage the flow cell itself. One
option here is to use our U-607.
This 100 psi back pressure regulator comes in a stainless steel holder,
so that the unit can provide outgassing protection and also serve
as a sinker to hold your tubing securely in the waste reservoir. Please
note: We highly recommend you contact the manufacturer of your detector
to determine the maximum pressure its flow cell can handle. If 100
psi is too much back pressure to add, please consider our back-pressure
regulators with lower pressure settings.
The use of secondary detection methods—such as fluorescence,
refractive index or mass spectrometry—also affects the proper
choice of back-pressure regulators. When secondary detection is employed,
our standard back-pressure regulators are often not the best choice,
as their internal volume is too great and may cause loss of sample
component resolution. In this instance, we recommend the use of our
Ultra Low Volume Back Pressure Regulator, specifically part number
M-415. This unit has a maximum internal volume of 6 uL,
and tests have shown that it does not significantly reduce the resolution
between sample components.
If your applications include the need to split the post-detector flow
into two separate pathways, (such as to a mass spectrometer and to
waste), then we would recommend the use of our P-451
Low Pressure Micro-Splitter Valve. This product is capable of taking
standard HPLC flow rates and splitting off a stream as low as 2 uL/minute.
The valve, which comes complete with the necessary fittings, is primarily
constructed of PEEK polymer and thus it offers excellent chemical
resistance. For an easier return to a previous setting, we recommend
our P-470,
Micro Splitter Valve, which comes with fittings and a built-in graduated
micrometer thimble for fine-tuning adjustments.
